After leaving high school, Wilkinson worked briefly as a dental assistant, and then as a glamour model.

Wilkinson met Hugh Hefner in 2004. She had been hired to be a nude body paint model at his 78th birthday party. After their first meeting he asked her to be one of his girlfriends and she moved into the Playboy Mansion.

Wilkinson starred, alongside Hefner's two other girlfriends, Holly Madison and Bridget Marquadt, in the reality television series The Girls Next Door. The series followed their lives living in the Playboy Mansion.

In 2009 she moved out of the Playboy Mansion, after she broke up with Hefner and met her future husband, Hank Baskett. Together they filmed her own reality show, Kendra, which centred around her living alone, and once she got engaged, planning her wedding.

Wilkinson married Baskett in June 2009. They have a son, Hank Baskett IV, born in December 2009.

In 2010 a sex tape of Wilkinson and an unidentified man was released.
